Specimen Collection Criteria

Collect: Two Lavender-top EDTA tubes OR two Yellow-top ACD tubes.

Invert several times to mix well.

Physician Office/Drawsite Specimen Preparation

Do not refrigerate or freeze specimen. Maintain whole blood at room temperature (20-26°C or 68-78.8°F) prior to transport. Transport to the Laboratory within 24 hours of collection.

Preparation for Courier Transport

Transport: Whole blood at room temperature (20-25°C or 68-77°F).

Rejection Criteria

  • Unlabeled specimens.
  • Specimens drawn in clot, SST, or Sodium Heparin tubes.

Clinical Utility

HLA Class II genotyping defines the antigens of the HLA-DRB1 and DQB1 loci of the Major Histocompatibility Complex found on the short arm of chromosome 6. Using DNA extracted from leukocytes, HLA-DRB1 and DQB1 alleles are amplified via PCR using sequence-specific primers. After amplification, the specific HLA antigens are determined by gel electrophoresis. HLA genotyping serves an important role in transplantation and disease association studies.
