Specimen Collection Criteria

Collect: Rectal biopsy for Hirschsprung Disease should be placed fresh (no formalin) in a specimen container on ice and delivered to Surgical Pathology immediately.

  • The specimen container must be labeled properly with patient's full name (first and last), date of birth, hospital number including visit, and specimen source.
  • The specimen must be accompanied by a Progress Notes Operative Form #5113, Anatomic Pathology Order Form and/or EPIC Anatomic Pathology Order requisition, including patient's full name, date of birth, patient's social security number (only last 4 of SSN), hospital number including visit number, surgeon and/or clinician name, specimen source including identifiers and pre-op diagnosis.

Rejection Criteria

  • Delay in processing of fresh (no formalin) rectal biopsy will affect the staining and diagnosis.
  • Any missing information will also delay processing and final diagnosis.

Inpatient Specimen Preparation

Fresh (no formalin) rectal biopsy should be delivered immediately to Surgical Pathology accompanied by Form #5113, Anatomic Pathology Order Form, and/or EPIC Anatomic Pathology Order requisition. The specimen should be handed directly to a Surgical Pathology assistant. Never leave a fresh rectal biopsy in the specimen pick-up area.

In-Lab Processing

The rectal biopsy for Hirschsprung Disease is immediately dictated; a portion of the biopsy is oriented on edge then frozen (-20°C/-4°F or below). The remainder of the tissue is submitted for processing, per Hirschsprung biopsy protocol. 

Storage

Specimen Stability for Testing:

Specimens must be transported to the Surgical Pathology Laboratory and processed immediately.

Specimen Storage in Department Prior to Disposal:

Frozen (-70°C/-94°F or below): 2 years

Royal Oak Surgical Pathology/Histology Laboratory.

Performed

Monday – Friday, 6:00 am – 3:00 pm.
Results available within 2-4 working days.

Test Methodology

Routine Hematoxylin and Eosin stain. Special staining procedures: Staining with, acetylcholinesterase. 

Interpretation

Microscopic diagnosis of prepared slides.
