Specimen Collection Criteria

Collect (preferred specimen): One Pink or one Lavender-top EDTA tube, or plain Red-top tube.

Also acceptable: Amniotic fluid, Cerebrospinal Fluid (CSF), Bone Marrow, Bronch Wash, Pericardial Fluid, Pleural Fluid, Trach Aspirate, Trach Wash, paraffin-embedded tissue, or fresh frozen tissue.

Specimen source is required.

Physician Office/Draw Specimen Preparation

  • Centrifuge to separate plasma or serum from cells. Transfer serum or plasma to a plastic transport tube and keep at room temperature (20-26°C or 68-78.8°F).
  • Maintain CSF, Bone Marrow, BAL, Bronch Wash, or Trach Aspirate specimens frozen (-20°C/-4°F or below) prior to transport.
  • Maintain Pleural fluid, Amniotic Fluid, or Pericardial Fluid at room temperature (20-26°C or 68-78.8°F).
  • Fresh tissue in sterile container must be frozen (-20°C/-4°F or below). Do not add water, saline or other fluid media to container. Paraffin-embedded tissue is not acceptable.

Preparation for Courier Transport

Transport: 2.0 mL serum, plasma, pericardial and pleural fluid, or amniotic fluid at room temperature (20-26°C or 68-78.8°F), Trach Aspirate, Trach Wash, Bronch Wash, BAL, Bone Marrow, CSF or fresh tissue, frozen (-20°C/-4°F or below). (Minimum: 0.5 mL)

Rejection Criteria

  • Heparinized specimens.
  • Specimens in non-sterile or leaking containers.
  • Specimens not collected and processed as indicated.

In-Lab Processing

  • Centrifuge to separate plasma or serum from cells. Transfer serum or plasma to a plastic transport tube and keep at room temperature (20-26°C or 68-78.8°F).
  • Maintain CSF, Bone Marrow, BAL, Bronch Wash, or Trach Aspirate specimens frozen (-20°C/-4°F or below) prior to transport.

Quantitative Polymerase Chain Reaction (PCR).

Clinical Utility

Parvovirus B19 manifests itself as an acute or chronic hematological disorder in immunocompromised patients. It can cause persistent anemia, sometimes associated with leukopenia and thrombocytopenia. Pediatric transplant patients are at risk for chronic infections, which can be associated with lung and/or renal disorders. Quantitative DNA PCR can be used to detect the presence of the virus, track the course of infection, and monitor response to treatment.
