Specimen Collection Criteria

Collect: One Gold-top SST tube. (Minimum Whole Blood: 2.0 mL)

Physician Office/Draw Specimen Preparation

Let specimen clot 30-60 minutes then centrifuge to separate serum from cells. Refrigerate (2-8°C or 36-46°F) the centrifuged collection tube within two hours of collection. (Minimum Serum: 0.5 mL)

Preparation for Courier Transport

Transport: Centrifuged collection tube, refrigerated (2-8°C or 36-46°F). (Minimum Serum: 0.5 mL)

Rejection Criteria

  • Plasma specimens.
  • Severely lipemic, icteric, or hemolyzed specimens. 

In-Lab Processing

Let specimen clot 30-60 minutes then immediately centrifuge to separate serum from cells. Room temperature is acceptable for a maximum of two hours. (Minimum Serum: 0.5 mL)

Test Methodology

Indirect Fluorescent Assay (IFA).

Interpretation

Titers in patients with primary biliary cirrhosis (PBC) range from 1:20 to 1:1280 with about 50% of PBC patients having titers between 1:200 and 1:1280.

Low anti-mitochondrial antibody (AMA) titers may be detected in various liver disorders such as chronic active hepatitis and cryptogenic cirrhosis. High AMA titers support the diagnosis of primary biliary cirrhosis. However, AMA titers do not appear to have any correlation with clinical activity since titers do not vary with the severity or progression of disease. Therefore, mitochondrial autoantibodies cannot be used to monitor responses to therapy or provide prognostic information.

Low titer positive results may occur in healthy individuals.

Clinical Utility

The anti-mitochondrial antibody assay aids in the diagnosis of primary biliary cirrhosis.
