Specimen Collection Criteria

Collect (preferred specimen): One plain Red-top tube.
Also acceptable: One Gold-top SST tube.

Send specimen for processing immediately after collection.

Physician Office/Draw Specimen Preparation

Critical frozen. Let specimen clot 30-60 minutes then centrifuge to separate serum from cells. Transfer serum to a plastic transport tube and freeze (-20°C/-4°F or below) immediately.

Preparation for Courier Transport

Transport: 1.0 mL serum, frozen (-20°C/-4°F or below). (Minimum: 0.15 mL)

Rejection Criteria

  • Grossly hemolyzed specimens.
  • Grossly lipemic specimens.
  • Specimens not collected and processed as indicated.

In-Lab Processing

Test Methodology

Latex Agglutination.

Interpretation

Any titer should be considered clinically significant, however, clinical correlation must be present.

Some cutaneous infections are associated with negative serologic results.

Extracutaneous infections, including disseminated and articular infections, produce positive tests.

Clinical Utility

This assay aids in detecting antibodies in extracutaneous sporotrichosis. Sporptrichosis usually is a subcutaneous mycosis caused by Sporothrix schenckii. The fungus typically is introduced by minor trauma into subcutaneous tissue and produces a mycotic infection. The infection may be limited to the site of inoculation or may spread along proximal lymphatic channels.
