Specimen Collection Criteria

Collect: Amniotic fluid in a sterile collection container. (Minimum: 7.0 mL)

Transport refrigerated specimen to the Laboratory for processing immediately after collection. 

Physician Office/Draw Specimen Preparation

Arrange for transportation to the Laboratory prior to collection. Do not centrifuge or freeze the specimen. Maintain refrigerated (2-8°C or 36-46°F) prior to transport.

Preparation for Courier Transport

Transport: Amniotic fluid, refrigerated (2-8°C or 36-46°F). (Minimum: 7.0 mL)

Rejection Criteria

Samples visibly contaminated with blood, meconium, or mucus.

Inpatient Specimen Preparation

Transport specimen to the Laboratory immediately after collection.

Storage

Specimen Stability for Testing:

Room Temperature (20-26°C or 68-78.8°F): Unacceptable
Refrigerated (2-8°C or 36-46°F): 72 hours
Frozen (-20°C/-4°F or below): Unacceptable

Specimen Storage in Department Prior to Disposal:

Refrigerated (2-8°C or 36-46°F): 30 days

Reference Range

Mature: Greater than or equal to 55,000/mcL.

Test Methodology

Sysmex XN Series.

Interpretation

Amniotic fluid Lamellar Body Count (LBC) greater than or equal to 55,000/mcL is predictive of fetal lung maturity. The main value of fetal lung maturity testing is in predicting the absence of respiratory distress syndrome (RDS).

Specimens with Lamellar Body Count (LBC) less than 55,000/mcL will not be sent out for L/S ratio testing.

Clinical Utility

The number of Lamellar Bodies in amniotic fluid reflects the degree of development of the fetal lung and the relative risk for neonatal respiratory distress if the fetus were to be delivered.
